Activin/Inhibins & Receptors

Activins and inhibins, belong to the TGF-β family superfamily, are structurally homologous but with functionally reverse actions. They were first described as regulators of follicle-stimulating hormone (FSH) secretion and erythropoiesis. Inhibin and activin share a common β-subunit, with activins occurring as a dimer of two β-subunits: activin A (βAβA), activin BA (βA βB), and activin B (B B ), whereas inhibins consist of an α-subunit and either a βA or a B-subunit. Activins are known to stimulate FSH release while inhibins inhibit it. Activins signal through heteromeric complexes of type II (ActRIIA, ActRIIB) and type I receptors (predominantly ActRIB, but also ActRIA and ActRIC). Activins bind to type II receptors and recruit type I receptors, which then phosphorylate the intracellular signaling proteins SMAD2/3. These assemble with SMAD4, translocate to the nucleus, and bind to specific target genes to modulate their expression. Inhibin also bind type II activin receptors but do not recruit ALK4 (type I receptors), providing a competitive model for the antagonism of activin by inhibin. Type III TGF-beta receptor, betaglycan, can function as an inhibin co-receptor with ActRII.
