Bone morphogenetic proteins (BMPs) are multi-functional growth factors that belong to the Transforming Growth Factor-β (TGF-β) superfamily of proteins. BMP were originally identified as osteoinductive components in extracts derived from bone. They are currently known to play important roles in a wide array of processes during formation and maintenance of various organs including bone, cartilage, muscle, kidney, and blood vessels. BMPs are typically secreted as homomeric dimers. They can signal through both canonical, Smad-dependent pathway and non-canonical pathways. In the canonical pathway, BMPs induce signaling through a heterotetrameric receptor complex composed of two dimers of type I and two type II serine-threonine kinase receptors, known as BMP receptors (BMPRs). The constitutively active type II receptor then transphosphorylates the type I receptor, and the type I receptor phosphorylates the R-Smads (Smad1/5/8). hosphorylated Smad1/5/8 associates with the co-Smad (Smad4), and the complex translocates to the nucleus where it further associates with coactivators or corepressors to regulate gene expression. Various non-canonical pathways, including the MAPK cascade, can also lead to regulation of gene expression. The BMPs play important roles in a multitude of processes during embryonic development and adult homeostasis by regulating cellular lineage commitment, morphogenesis, differentiation, proliferation, and apoptosis of various types of cells throughout the body.
