CD160 is an Ig-like, glycosylphosphatidylinositol (GPI)-anchored cell membrane protein expressed on cytotoxic NK and CD8+ T cells, NKT cells, a minority of CD4+ T cells, γδ T cells, intraepithelial lymphocytes (IELs), intraepithelial innate lymphoid cell type 1 (ILC1) cells, and mast cells. CD160 belongs to the Ig superfamily. CD160 binds to MHC class I molecules with low affinity and to herpes virus entry mediator (HVEM), a TNF receptor family member, with much higher affinity. Both co-inhibitory and co-stimulatory functions for CD160 have been reported. On the one hand, CD160 can stimulate NK cytotoxicity and IFN-γ production and enhance T cell activity. Such effects have been attributed to the ability of CD160 to bind classical and nonclassical MHC class I molecules. On the other hand, CD160 acts as a T cell co-inhibitory molecule that interacts with the HVEM on antigen-presenting cells to provide an inhibitory signal to T cells.
