CD96 & CD155/CD111/CD112

CD96 (also known as TACTILE) is a member of the immunoglobulin (Ig) superfamily whose expression mainly by cells of hematopoietic origin, in particular on T and NK cells. CD96 may play a role in the adhesive interactions of activated T and NK cells during the late phase of the immune response. CD96 is a promising candidate for immunotherapy and plays an important role in anti-tumor immune responses. CD96, TIGIT and CD226 all share a common ligand with strikingly different affinities, CD155 (poliovirus receptor, PVR), and belong to the PVR-nectin family. CD96 also binds to CD111. Similar to the CD28/CTLA-4 pathway, CD96 and TIGIT act as co-inhibitory receptors together with the co-stimulatory receptor CD226, in which shared ligands and differential receptor: ligand affinities fine-tune the immune response. In addition, CD96 promotes NK cell-target cell adhesion and inhibits cytokine responses induced by NK cells by competing with CD226 for CD155 binding. A further found that CD96 blockade can be more effective when combined with other therapies including anti-CTLA-4, anti-PD-1, or doxorubicin chemotherapy. Interestingly, recent study suggests CD96 functions as a co-stimulatory receptor to enhance CD8+ T cell activation and effector responses.
