Transforming growth factor beta (TGF-β) is a small (25 kDa), highly pleiotropic cytokine that regulates cell growth and differentiation, apoptosis, cell motility, extracellular matrix production, angiogenesis, and cellular immune responses. Five TGF-β isoforms have been identified with three (TGF-β1, TGF-β2, and TGF-β3) present in mammals. The amino acid sequences of the three isoforms are 70-80% homologous. TGF-β1 is expressed in endothelial, hematopoietic, and connective tissue cells; TGF-β2 in epithelial and neuronal cells; and TGF-β3 primarily in mesenchymal cells. All three TGF-βs use the same receptor and it has three components: two transmembrane serine/threonine kinase receptors, TβR-I and TβR-II, and a co-receptor, TβR-III. TβR-III is the most abundant receptor type, but it is not involved in signaling. TβR-III binds TGF-β (all isoforms) and recruits TGF-β to TβR-II. Once TβR-II is engaged, TGF-β results in the formation of a TβR-I/TβR-II heteromeric complex, in which TβR-II phosphorylates TβR-I and thus activates TβRI. Subsequently, the activated TβRI recruits and phosphorylates the R-Smad proteins, Smad2 and Smad3, and the latter form a heteromeric complex with Smad4, which translocates to the nucleus, binds to DNA and regulates transcription. TGF-βs exert multiple effects on inflammation, angiogenesis, fibrosis and tumor progression. In addition, their importance is demonstrated by the many human diseases that result from disruption or dysregulation of the TGF-β signaling pathway, including developmental disorders, such as Marfan’s disease, and adult diseases such as cancer and fibrosis.
