Receptor serine/threonine kinases (RSTKs) are transmembrane proteins of the plasma membrane and are characterized by extracellular ligand-binding domains and cytoplasmic kinase domains. RSTKs mediate inhibitory as well as stimulatory signals for growth and differentiation by binding to members of the transforming growth factor-β (TGF-β) superfamily, e.g. TGF-beta and activins. TGF-β family receptors are grouped into three types, type I, type II, and type III. TβRIII functions as a co-receptor to increase ligand binding to TβRII. Once bound to TGF-β, TβRII recruits and binds TβRI, and transphosphorylates threonine and serine residues in the TTSGSGSG motif of TβRI, thereby stimulating its protein kinase activity. In the Smad pathway, the activated TβRI phosphorylates transcription factors Smad. The resulting Smad complex translocates into the nucleus and interacts with other transcription factors to specifically regulate the transcription of a multitude of TGF-β-responsive genes. There are seven type I receptors (also called as activin receptor-like kinases, ALKs) in mammals, ACVRL1 (ALK1), ACVR1 (ALK2), BMPR1A (ALK3), ACVR1B (ALK4), TGFBR1 (ALK5), BMPR1B (ALK6), and ACVR1C (ALK7). Among them, ALK5 can mediate TGF-β signaling with the TβRII to activate Smad2/3 in universal cell types, while in endothelial cells ALK1 functions with TβRII to activate Smad1/5/8 for TGF-β signaling. ALK2/3/6 can activate Smad1/5/8 with the type II receptors BMPRII, ActRII and ActRIIB in response to BMPs. ALK4/7 can activate Smad2/3 with ActRII and ActRIIB to mediate activin/Nodal signaling.
