Background

MST3/STK24 Protein, a serine/threonine-protein kinase, plays a pivotal role in apoptosis initiation in response to stress stimuli and caspase activation. Under oxidative stress conditions, it mediates cell death by modulating the phosphorylation of JNK1-JNK2 and p38 MAPKs, highlighting its involvement in stress-induced apoptotic pathways. Additionally, MST3/STK24 contributes to a caspase-independent apoptotic pathway by regulating the nuclear translocation of AIFM1 and ENDOG, as well as the DNase activity associated with ENDOG during staurosporine-induced apoptosis. The kinase activity extends to phosphorylating STK38L and stimulating its function. In collaboration with STK26, MST3/STK24 negatively regulates Golgi reorientation during polarized cell migration induced by RHO activation. Furthermore, it influences cellular migration by altering PTPN12 activity and PXN phosphorylation, inhibiting PTPN12 and potentially regulating PXN phosphorylation. Notably, MST3/STK24 may act as a key regulator in axon regeneration, particularly in the optic nerve and radial nerve, emphasizing its diverse roles in cellular processes.
