Independently, Gankyrin regulates EGF-induced AKT activation by inhibiting the RHOA/ROCK/PTEN pathway. It plays a key role in RAS-induced tumorigenesis, acting as a proto-oncoprotein by negatively regulating RB1 and p53/TP53. Gankyrin overexpression leads to RB1 phosphorylation and degradation, while also competing with CDKN2A for CDK4 binding. Additionally, Gankyrin facilitates MDM2-p53/TP53 binding and promotes p53/TP53 ubiquitination by MDM2, influencing the TP53:MDM2 complex's targeting to the 26S proteasome.
