KLD-12 is a 12-residue self-assembling peptide that is used in tissue-engineering. KLD-12 combined with SDF-1 self-assembled polypeptide enhances chondrogenic differentiation of bone marrow stromal cells (BMSCs). KLD-12 hydrogel can fill full-thickness osteochondral defects in situ and improve cartilage repair[1][2][3].

In Vitro

KLD-12 forms self-assembled polypeptide with SDF-1, improves the survival of bone marrow stromal cells (BMSCs), promotes osteogenic differentiation of BMSCs and cell migration through Wnt/β-catenin pathway[1].

In Vivo

KLD-12 (10-250 μg/rat, single dose, injection to infracture site) promotes bone regeneration at the fracture site in a dose-dependent manner in Sprague-Dawley rats model with drill-hole injury in femur[2].

1.  Calculate the length of the peptide.

2.  Calculate the overall charge of the entire peptide according to the following table:

2.  If water fails, add NH4OH (<50 μL).
3.  If the peptide still does not dissolve, add DMSO (50-100 μL) to solubilize the peptide.
Positive (>0) 1.  Try to dissolve the peptide in water first.
2.  If water fails, try dissolving the peptide in a 10%-30% acetic acid solution.
3.  If the peptide still does not dissolve, try dissolving the peptide in a small amount of DMSO.
Zero (=0) 1.  Try to dissolve the peptide in organic solvent (acetonitrile, methanol, etc.) first.
2.  For very hydrophobic peptides, try dissolving the peptide in a small amount of DMSO, and then dilute the solution with water to the desired concentration.
