LRP-5 Protein, Human (HEK293, mFc)

Cat. No.: HY-P77989
COA Handling Instructions

LRP-5 Protein functions as a coreceptor, collaborating with frizzled receptors to initiate the canonical Wnt signaling pathway. It crucially regulates cell fate determination, self-renewal, and plays a significant role in posterior epiblast patterning during gastrulation and bone development by influencing osteoblast proliferation and differentiation. LRP-5 facilitates the formation of a signaling complex, stabilizing beta-catenin and activating TCF/LEF-mediated transcription. As a coreceptor for non-Wnt proteins like norrin/NDP, LRP-5 contributes to retinal vascular development. Interactions with partners, including FZD8, AXIN1, DKK1, MESD, KREMEN2, CSNK1E, SOST, APCDD1, and CAPRIN2, underscore its intricate involvement in diverse cellular processes. LRP-5 Protein, Human (HEK293, mFc) is the recombinant human-derived LRP-5 protein, expressed by HEK293, with C-mFc labeled tag. The total length of LRP-5 Protein, Human (HEK293, mFc) is 620 a.a., with molecular weight of 110-115 kDa.

Background

LRP-5 protein acts as a pivotal coreceptor, partnering with the frizzled family of seven-transmembrane spanning receptors to transduce signals initiated by Wnt proteins. This activation leads to the canonical Wnt signaling pathway, a critical regulator of cell fate determination and self-renewal during both embryonic development and adult tissue regeneration. LRP-5 is particularly implicated in the posterior patterning of the epiblast during gastrulation and plays a crucial role in bone development by regulating osteoblast proliferation and differentiation. Mechanistically, LRP-5 facilitates the formation of the signaling complex between Wnt ligands, frizzled receptors, and itself, leading to the recruitment of AXIN1. This stabilizes beta-catenin/CTNNB1 and activates TCF/LEF-mediated transcriptional programs. Moreover, LRP-5 serves as a coreceptor for non-Wnt proteins, such as norrin/NDP, with a demonstrated role in retinal vascular development. The interaction with various partners, including FZD8, AXIN1, DKK1, MESD, KREMEN2, CSNK1E, SOST, APCDD1, and CAPRIN2, highlights its intricate involvement in the regulation of diverse cellular processes.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

  • Dilution Calculator

  • Specific Activity Calculator

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
= ÷

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
