Tapasin Protein, Human (HEK293, His)

Cat. No.: HY-P73619
COA Handling Instructions

Tapasin protein is key to MHC class I assembly, binding to TAP and forming MHC class I with peptides. As a heterodimer of PDIA3, tapasin mediates interactions between MHC class I, calreticulin, PDIA3, and TAP. Tapasin Protein, Human (HEK293, His) is the recombinant human-derived Tapasin protein, expressed by HEK293 , with C-His labeled tag. The total length of Tapasin Protein, Human (HEK293, His) is 394 a.a., with molecular weight of 45-50 kDa.

Tapasin Protein, Human (HEK293, His) is the recombinant human-derived Tapasin protein, expressed by HEK293 , with C-His labeled tag. Background

Tapasin protein assumes a pivotal role in the intricate processes of MHC class I assembly, specifically in the association with transporter associated with antigen processing (TAP) and the formation of MHC class I with peptides during peptide loading. Forming a heterodimer with PDIA3 through disulfide linkage, tapasin becomes an obligatory mediator in the interaction among newly assembled MHC class I molecules, calreticulin, PDIA3, and TAP. Notably, up to four MHC class I/tapasin complexes bind to a single TAP, as documented in various studies. Furthermore, tapasin exhibits an additional interaction with the HLA-G-B2M complex, a critical engagement that is essential for the loading of high-affinity peptides, elucidating its multifaceted role in the orchestration of MHC class I antigen presentation.

Molecular Weight

Approximately 45-50 kDa due to the glycosylation.

Purity

Greater than 90% as determined by reducing SDS-PAGE.

Appearance

Lyophilized powder.

Formulation

Lyophilized from a 0.2 μm filtered solution of PBS, pH 7.4.

Endotoxin Level

<1 EU/μg, determined by LAL method.

Reconstitution

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH2O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Storage & Stability

Stored at -20°C for 2 years. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.

Shipping

Room temperature in continental US; may vary elsewhere.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

  • Specific Activity Calculator

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
= ÷

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
