Frizzled-7

Frizzled-7 (FZD7) is one of the most broadly expressed members of the family and is unusual among Frizzled receptors for responding to multiple canonical Wnt ligands, including Wnt3a, Wnt8 and Wnt11. Ligand binding to its CRD activates beta-catenin-dependent transcription through Dishevelled and the destruction-complex axis, and it can also engage JNK-driven planar cell polarity responses. FZD7 is highly expressed in proliferative and stem cell compartments of intestine, skin and other epithelia. In cancer, FZD7 is frequently upregulated in hepatocellular carcinoma, esophageal and cervical tumors, where it sustains Wnt-driven self-renewal of cancer stem cells, and FZD7-targeted antibodies have shown tumor-suppressive activity in preclinical models, underscoring its drug-development relevance.
