ErbB2/HER2

ErbB2, universally known as HER2, is an orphan receptor tyrosine kinase of the ERBB family that has no known direct ligand and instead serves as the preferred heterodimerization partner for other ERBB receptors, amplifying their signaling. Gene amplification and protein overexpression occur in roughly 15-20% of breast cancers and define an aggressive subtype sensitive to trastuzumab, pertuzumab, and the antibody-drug conjugate trastuzumab deruxtecan. HER2 is also overexpressed in gastric and gastroesophageal cancers. Its extracellular domain dimerizes stably, making recombinant HER2 protein a standard reagent for antibody characterization and CAR-T development.
