Description & Advantages

MCE Mouse Fetal Brain (Differentiation) Organoid Kit includes a basic culture medium and differentiation supplements, designed for the induction and culture of mouse fetal brain organoids (mFBs).

During the expansion phase, mFBs are primarily composed of neural stem cells characterized by the expression of stem cell markers such as Sox2. Upon switching from the expansion medium to the differentiation medium, mFBs undergo differentiation into mature neuronal cells, exhibiting the expression of lineage-specific markers including Gfap, Map2, Cux1, and Dcx.

Protocol

Preparation of Mouse Fetal Brain Organoid Differentiation Medium

Prepare the differentiation medium according to the table below, mix thoroughly, and keep on ice. Perform all procedures under sterile conditions.

Differentiation of Mouse Fetal Brain Organoids

1. Initial expansion culture: Culture the organoids with the corresponding Mouse Fetal Brain Organoid (Expansion) Kit (MCE Cat. No.: HY-K6143) for at least 2-5 d to support healthy growth and tissue integrity.

2. Medium pre-warming: Pre-warm the differentiation medium to 37°C before use.

3. Organoid washing: Carefully remove the expansion medium and wash the organoids once with Basal Medium A.

4. Distribution and medium addition: Transfer 4-8 organoids into each well of a 6-well plate and carefully add pre-warmed differentiation medium. Avoid bubbles and excessive mechanical stress.

5. Differentiation culture: Continue culture for 7-9 d and completely replace the medium with fresh pre-warmed differentiation medium every other day.

6. Completion: At the end of the differentiation period, the organoids display neuronal morphology and differentiation characteristics and are ready for downstream neuron-marker analysis.
