Density Gradient Medium, also known as Iodixanol, is a low osmolality, non-ionic, iodinated density gradient medium.

MCE Density Gradient Medium (Iodixanol) is a sterile and endotoxin-tested 60% (w/v) aqueous solution of iodixanol for in vitro isolation of biological particles. It is commonly used for the isolation of molecules, cells and subcellular organelles including DNA, cytokines, lysosomes, endoplasmic reticulum, peroxisomes, T-lymphocytes and natural killer cells.

Protocol

Preparation and Application of Density Gradients

1. The product is a 60% (W/V) aqueous iodixanol solution. Prepare continuous, discontinuous, or self-forming density gradients using an appropriate buffer according to the density required for the target particles or cells.

2. Select a suitable dilution system for the separation target and prepare each density layer separately, mixing thoroughly.

3. For a discontinuous density gradient, carefully layer the solutions from highest to lowest density along the wall of the centrifuge tube and minimize mixing between layers.

4. Gently load the sample onto the density gradient and apply the centrifugation conditions appropriate for the target cells, viruses, organelles, or macromolecules.

5. After centrifugation, collect the target interface or density fraction and wash or exchange buffer as required for downstream applications.

Note: The product density is 1.320 ± 0.001 g/mL and endotoxin is ≤ 1 EU/mL. Working density, number of layers, and centrifugation conditions should be selected according to the target material and experimental protocol.
