Leibovitz's L-15 Medium
MCE Leibovitz's L-15 Medium (L-15 Medium) is a cell culture medium specifically designed for use without supplemental CO₂. Unlike conventional media that rely on a sodium bicarbonate–CO₂ buffering system, L-15 Medium uses phosphates and free-base amino acids to buffer pH. It can therefore maintain the acid–base balance of the culture without supplemental CO₂ and support stable cell growth under atmospheric conditions.

MCE Leibovitz's L-15 Medium (L-15 Medium) is a cell culture medium specifically designed for use without supplemental CO₂. Unlike conventional media that rely on a sodium bicarbonate–CO₂ buffering system, L-15 Medium uses phosphates and free-base amino acids to buffer pH. It can therefore maintain the acid–base balance of the culture without supplemental CO₂ and support stable cell growth under atmospheric conditions.
Originally developed by Leibovitz for cell culture without sodium bicarbonate or supplemental CO₂, this medium is now widely used to maintain established cell lines and to isolate and culture primary cells from human and animal embryonic and adult tissues (such as Hep-2 cells).

Protocols
L-15 Medium is a basal medium that does not contain proteins, lipids, or growth factors. Add appropriate supplements according to the cell type and experimental requirements. For routine culture, approximately 10% fetal bovine serum (Cat. No.: HY-T1000) may be added. For low-serum or serum-free culture, supplements such as insulin-transferrin-selenium (ITS) (Cat. No.: HY-K3018) may be added as needed.
This product contains L-Alanyl-L-Glutamine rather than free L-Glutamine added directly. Free L-Glutamine gradually degrades during storage in liquid medium, producing ammonia and pyroglutamic acid, which can be harmful to cells. In contrast, L-Alanyl-L-Glutamine is stable in aqueous solution and does not spontaneously degrade. During culture, peptidases released by cells gradually hydrolyze it into L-Alanine and L-Glutamine for sustained cellular use. This can improve nutrient availability, reduce ammonia accumulation, and extend the storage stability of the medium.
Note: a. This product does not contain antibiotics. Penicillin-Streptomycin Solution (Cat. No.: HY-K1006) may be added if required by the experiment. After adding serum, ITS, antibiotics, or other supplements, store the prepared medium at 2–8°C and use it within one week.
b. This product is designed for use without supplemental CO₂. Maintain CO₂-free conditions during use and supplementation; no additional CO₂ adjustment is required.
Application
MCE Leibovitz's L-15 Medium is suitable for culturing the established Hgill-W1 cell line and primary explants from human and animal embryonic and adult tissues without supplemental CO₂. It can be used for cell transport, microscopic observation and manipulation, maintenance of cell growth and viability, and research and production applications involving tissues and cells.

Attention
1. This product has been sterilized by filtration. Use aseptic technique to avoid contamination.
2. Because peptide hydrolysis and dipeptide utilization take time, cells cultured in this medium may have a slightly longer lag phase. This is normal.
3. This product is designed for use without CO₂. Use it in a temperature-controlled incubator without supplemental CO₂. Do not place it in a CO₂ incubator, as the absence of bicarbonate buffering may cause the medium to become excessively acidic and impair cell growth and viability.
4. This product is intended for scientific research by trained personnel only. It must not be used for clinical diagnosis or treatment, or in food or pharmaceutical products.
5. For your safety and health, wear a lab coat and disposable gloves when handling this product.
