Description & Advantages

The Bradford protein concentration assay is recognized as one of the most sensitive protein concentration assays available. The principle is that Coomassie Brilliant Blue G250 exhibits a red color under acidic conditions, when it binds with basic amino acids (predominantly arginine) or aromatic amino acids within proteins, the solution undergoes a color change from red to blue, with the absorbance maximum moving from 465 nm to 595 nm, and the color intensity is directly proportional to the protein content present. By measuring the absorbance of a standard protein, a rapid detection of thee protein concentration can be achieved.

Protocol

1. Preparation of BSA standards

Use the supplied Standard Protein Solution (5 mg/mL BSA) to prepare a series of BSA standards. A recommended standard range is shown below:

2. Microplate method

1. Add 20 μL BSA standard or protein sample to each well of a 96-well plate. Dilute samples in advance if their concentration is outside the standard range.

2. Add 200 μL G250 Staining Solution to each well.

3. Mix gently and incubate for approximately 5 min at room temperature.

4. Measure absorbance at 595 nm, preferably within 30 min after addition of the staining reagent.

3. Calculation of protein concentration

Plot BSA concentration versus A595, calculate sample protein concentration from the standard curve, and multiply by the corresponding dilution factor.

Note: Bradford assays are sensitive to detergents and other interfering components. Match the sample buffer and standard matrix whenever possible and include an appropriate blank.
