Western blot analysis was performed on extracts from SH-SY5Y (lane 1, 15 μg), U-87MG (lane 2, 15 μg), 3T3 (lane 3, 15 μg), C2C12 (lane 4, 15 μg), and C6 (lane 5, 15 μg) using PDGFR beta Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Actin, HY-P80438, 1:20000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.

PDGFR beta is a PDGF Receptor beta is a tyrosine-protein kinase that functions as a cell-surface receptor for homodimeric PDGFB and PDGFD, as well as heterodimers formed by PDGFA and PDGFB. It plays essential roles in embryonic development, cell proliferation, survival, differentiation, chemotaxis, and migration. Critical for blood vessel development, it promotes the proliferation, migration, and recruitment of pericytes and smooth muscle cells to endothelial cells. It also contributes to vascular smooth muscle cell migration and neointima formation at injury sites, and is required for normal cardiovascular system development, pericyte (mesangial cell) recruitment in kidney glomeruli, and glomerular capillary network formation. The receptor induces actin cytoskeleton rearrangement and membrane ruffle formation. Ligand binding activates multiple signaling cascades, phosphorylating PLCG1, PIK3R1, PTPN11, RASA1/GAP, CBL, SHC1, and NCK1, thereby regulating downstream effectors such as diacylglycerol, inositol 1,4,5-trisphosphate, Ca2+ mobilization, protein kinase C, AKT1, HRAS, RAF1, and MAPK1/ERK2/MAPK3/ERK1. Receptor signaling is downregulated by phosphatase-mediated dephosphorylation and rapid internalization. by similar: It also promotes phosphorylation and activation of SRC family kinases, PDCD6IP/ALIX, and STAM.
