MCE Medium 199 (with Earle's Balanced Salts), also known as Medium 199 (M199), was originally developed by Morgan, Morton, and Parker in 1950 to study the nutritional requirements of chick embryo fibroblasts. It is supplied as a ready-to-use liquid medium. The formulation contains a broad range of vitamins, amino acids, and other defined components and was initially used to investigate the nutritional requirements for the growth of primary explant cells under chemically defined conditions.

MCE Medium 199 (with Earle's Balanced Salts), also known as Medium 199 (M199), was originally developed by Morgan, Morton, and Parker in 1950 to study the nutritional requirements of chick embryo fibroblasts. It is supplied as a ready-to-use liquid medium. The formulation contains a broad range of vitamins, amino acids, and other defined components and was initially used to investigate the nutritional requirements for the growth of primary explant cells under chemically defined conditions.

Compared with DMEM, M199 contains relatively lower concentrations of basal salts, vitamins, and amino acids. It also contains folic acid and inositol. M199 is suitable for cells from a wide range of species, particularly undifferentiated cells and primary cell cultures. It is especially suitable for the in vitro culture of primary explants such as mouse embryonic epithelial cells and rat choroid plexus tissue.

Protocols

Compared with other basal media, MCE M199 contains distinctive components such as adenine, adenosine, hypoxanthine, thymine, and various vitamins. As a basal medium, M199 does not contain proteins, lipids, or growth factors. Add appropriate supplements according to the cell type and experimental requirements. For routine culture, approximately 10% fetal bovine serum (Cat. No.: HY-T1000) may be added. For low-serum or serum-free culture, supplements such as insulin-transferrin-selenium (ITS) (Cat. No.: HY-K3018) may be added as needed.

This product contains L-Glutamine, which gradually degrades during storage in liquid medium. If the medium has been stored at 2–8°C for an extended period, assess whether additional L-Glutamine is needed based on the cells’ requirements and culture performance.

Note: This product does not contain antibiotics. Penicillin-Streptomycin Solution (Cat. No.: HY-K1006) may be added if required by the experiment. After adding serum, ITS, antibiotics, or other supplements, store the prepared medium at 2–8°C and use it within one week.

Application

MCE M199 is widely used in virology research, vaccine production, and the in vitro culture of various animal cells, particularly primary cells and tissues. It is a well-established medium for research and biomanufacturing applications.

Attention

1. This product has been sterilized by filtration. Use aseptic technique to avoid contamination.

2. This product is formulated with Earle's salts. Earle's salts contain a bicarbonate buffering system and are suitable for use in a 5%–10% CO₂ incubator. In contrast, Hanks' salts have lower buffering capacity and are designed for culture without supplemental CO₂, including open-system culture outside a CO₂ incubator.

3. This product is intended for scientific research by trained personnel only. It must not be used for clinical diagnosis or treatment, or in food or pharmaceutical products.

4. For your safety and health, wear a lab coat and disposable gloves when handling this product.
