MCE Bi-27 Serum-free Supplement (50×), Antioxidant-Free is a customized neuronal culture supplement developed by removing antioxidants components (such as vitamins, superoxide dismutase, catalase, etc.) from the standard Bi-27 formulation. It is specifically designed for investigating the roles of reactive oxygen species (ROS) in aging, cytotoxicity, apoptosis, and chronic neurological diseases. This formulation provides a stable culture environment for mechanistic studies without the interference of exogenous antioxidants.

    Immunofluorescence (IF) analysis of SD rat cortical neurons: After 7 d of in vitro culture (10× magnification), three culture conditions were compared: no Bi-27 supplement (W/O), Bi-27 Serum-free Supplement (50×), Antioxidant-Free (HY-K3014D), and Gibco B-27 Supplement, minus antioxidants (Competitor). The results showed that supplementation with B-27 significantly improved neuronal adhesion, increased cell density, and promoted more extensive and structurally developed neurite outgrowth. In contrast, the W/O group exhibited poor overall cell condition, characterized by weak attachment, reduced cell numbers, and limited neurite development. Further comparison indicated that HY-K3014D and Gibco B-27 Supplement, minus antioxidants (Competitor) performed comparably in supporting the growth and differentiation of SD rat cortical neurons, with no statistically significant differences were observed between the two groups. Overall, MCE HY-K3014D demonstrated performance equivalent to the imported competitor in maintaining neuronal morphology, promoting cell growth, and supporting neurite development.

    Analysis of CTG assay and microscopic observation on intestinal cancer cells: After 6 days of in vitro culture, three culture systems including no Bi-27 supplement (W/O), Bi-27 Serum-free Supplement (50×), Antioxidant-Free (HY-K3014D), and Gibco B-27 Supplement, minus antioxidants (Competitor) were compared. Comprehensive evaluation of cell growth status was conducted via microscopic observation (4×, 10×, 20× magnification) and CellTiter-Glo (CTG) luminescence detection. The results showed that compared with the no-Bi-27 control (W/O), supplementation with insulin-free Bi-27 improved overall cell growth, as evidenced by more intact cellular structures, clearer organoid/cell cluster morphology, and a trend toward increased size. CTG quantification further indicated no statistically significant differences (ns) in cell viability among the groups. In particular, HY-K3014D and Gibco B-27 Supplement, minus antioxidants (Competitor) exhibited comparable performance in supporting cell viability, with neither showing a significant difference relative to the control. In summary, within this colorectal cancer cell culture system, HY-K3014D Bi-27 demonstrated performance equivalent to the imported competitor in supporting cell growth and maintaining cell viability, with no significant differences observed.

    MCE Bi-27 Serum-free Supplement (50×), Antioxidant-Free is a customized neuronal culture supplement developed by removing antioxidants components (such as vitamins, superoxide dismutase, catalase, etc.) from the standard Bi-27 formulation. It is specifically designed for investigating the roles of reactive oxygen species (ROS) in aging, cytotoxicity, apoptosis, and chronic neurological diseases. This formulation provides a stable culture environment for mechanistic studies without the interference of exogenous antioxidants.

    The recommended working concentration of Bi-27 Supplement is 2% (v/v), corresponding to a 1:50 dilution before use in the culture medium.

    Protocol

    Prepare complete Neuronal basal Medium

    1. Thaw MCE Bi-27 Serum-free Supplement (50×), Antioxidant -Free overnight at 4°C.

    2. Aseptically add 2% Bi-27 and 0.5mM L-Glutamine to the Neuronal Basal Medium before use.

    3. Primary Hippocampal Neuron Culture: Add 25 µM of L-glutamate to the complete Neuronal Basal Medium. The medium can be replaced after 4 days of culture, and glutamate should not be added to the medium after this change.

    Note: Once prepared, the complete Neuronal Basal Medium can be stored at 4°C for two weeks, protected from light.

    Attention

    1. This product appears as a light yellow liquid. The color may not be consistent following freezing, but it can fully dissolve in a 37°C water bath.

    2. It is recommended to appropriately aliquot the solution to avoid repeat freeze-thaw cycles.

    3. Pay attention to aseptic operation to avoid contamination.
