PVRIG (CD112R) is an inhibitory immune-checkpoint receptor that binds only to its ligand CD112 (Nectin-2) with high affinity on target cells suppressing lymphocyte cytotoxic function. CD112R is expressed in CD4+ and CD8+T cells, γδ T, NKT, and NK cells. CD112R interacts with CD112 competing with DNAM-1 but not with CD155 (PVR). It has been demonstrated that blocking the CD112-CD112R axis increases effector T-cell function in cancer, suggesting that the CD112-CD112R and CD155-TIGIT pathways could be non-redundant inhibitory signaling pathways.
