Description & Advantages
The lentiviral vector is a gene therapy vehicle derived from HIV-1, capable of stably integrating exogenous genes into the host cell genome. It features a broad host range, enables efficient infection of both dividing and non-dividing cells, and ensures long-term and stable expression of transgenes. Therefore, it has become a powerful tool for gene delivery, widely used in gene overexpression, RNA interference, microRNA studies, and in vivo animal experiments across various cell lines.
MCE Lentivirus Concentration Solution (5×) is a high-efficiency polymer solution optimized for lentiviral particle enrichment, providing a simple, rapid, and effective method for lentivirus concentration. To use, simply mix the lentiviral supernatant with the reagent at the recommended ratio, incubate briefly, and centrifuge using a standard laboratory centrifuge to obtain a visible lentiviral pellet.
Features of MCE Lentivirus Concentration Solution (5×)

Protocol
1. Pretreatment of Viral Supernatant
Collect lentiviral culture supernatant and remove cells and large debris by low-speed centrifugation. Filter through a 0.45 μm membrane if required.
2. Concentration
1. Completely thaw Lentivirus Concentration Solution (5×) and mix gently.
2. Add 1 part Lentivirus Concentration Solution (5×) to 4 parts viral supernatant to obtain a final 1× concentration.
3. Mix gently and incubate at 4°C for several hours or overnight to allow viral particles to precipitate.
4. Centrifuge at approximately 4,000 × g for approximately 30 min at 4°C.
5. Carefully remove the supernatant without disturbing the viral pellet.
6. Resuspend the viral pellet in an appropriate volume of sterile PBS, culture medium, or Lentivirus Solution Buffer. The virus may be concentrated to approximately 1/10 of the original volume as required.
Note: Aliquot concentrated lentivirus and store at -80°C. Avoid repeated freeze-thaw cycles. MCE validation demonstrated approximately 10-fold concentration of viral supernatant.
