IL-4 Receptor (IL-4R) is a receptor for IL-4 and IL-13 and belongs to the type I cytokine receptor family, subfamily 4. IL-4R binds to the JAK1/2/3 STAT6 pathway and participates in the response to promote Th2 cell differentiation by binding to IL-4. Soluble IL4R (sIL4R) inhibits IL4-mediated cell proliferation and T cell up-regulation of IL-5[1][2]. Among them, IL4RA is the main functional chain of IL-4R, a receptor of B cell stimulator (BSF) and an anti-inflammatory cytokine, which is highly expressed in activated T cells[3]. IL-4 RA is involved in the formation of two interleukin receptors in different cell types. The formation of type I receptor depends on the recruitment of IL-2Rγ chain by IL-4 RA and then binding to IL-4. The formation of type II receptor depends on the recruitment of IL-13Rα-1 chain by IL-4 RA to bind IL-4[4][5]. IL-4 RA produces a soluble form through alternate splicing or proteolysis that preserves ligand binding properties and inhibits IL-4 biological activity. The soluble subtype of IL-4 RA is produced by metalloproteinases on the cell surface through lyolytic cleavage (abscission)[6]. IL-4 RA plays an important role in Th2-biased immune response, replacement macrophage activation, mucosal immunity, allergic inflammation, tumor progression, and atherosclerosis formation[7].
