MCE Gentamicin-Amphotericin B (500×), sterile is used to prevent contamination of cell culture due to their effective combined action against gram-positive bacteria, gram-negative bacteria, fungi and yeasts.
MCE Gentamicin-Amphotericin B (500×) is a liquid formulation which contains 5 mg/mL of gentamicin, and 125 μg/mL of amphotericin B in 0.85% NaCl. Common working concentration for gentamicin is 10 μg/mL, and for amphotericin B is 0.25 μg/mL.
Protocol
General Protocol
1. This product is a sterile 500× stock containing Gentamicin at 5 mg/mL and Amphotericin B at 125 μg/mL for supplementation of cell culture media.
2. Completely thaw and mix gently. Under sterile conditions, add at 1 : 500, i.e. 2 μL per 1 mL medium or 2 mL per 1 L medium.
3. At 1× working concentration, the final concentrations are 10 μg/mL Gentamicin and 0.25 μg/mL Amphotericin B.
4. Mix thoroughly after addition and use for routine mammalian cell culture. The working concentration may be optimized according to cell type, contamination-control requirements, and experimental purpose.
Note: This is a sterile solution and should be handled aseptically. Store at -20°C, avoid repeated freeze-thaw cycles, and aliquot for routine use when appropriate.
