MCE ECM Gentle Dissociation Solution is a gentle ECM-degrading enzyme mixture derived from marine bacteria and Bacillus species, specifically formulated for efficient and low-damage digestion of in-vitro cell systems. It selectively degrades extracellular matrix components while minimizing disruption to the cell membrane and intercellular junctions, thereby significantly reducing mechanical stress during dissociation.

This product is compatible with a wide range of cell types, including stem cell colonies, primary cells, neural cells, and organoids, and is particularly well suited for gentle yet effective dissociation of brain organoids and other complex 3D structures. Compared with conventional proteases or trypsin, this mild dissociation solution provides more precise control over the digestion process, better preserves cell viability and functionality, and serves as an ideal tool for stem cell research, organoid culture, and regenerative medicine applications.

Storage

Cardiomyocyte Dissociation Reagent A: 4°C, 2 years.

Cardiomyocyte Dissociation Reagent B: -20°C, 2 years. Avoid repeated freeze/thaw cycles.

Protocol

For detailed procedures, please refer to the Replating and Confocal Imaging Identification section of the Human iPSC/ESC Cardiomyocyte Induction Differentiation Kit (Cat. No.: HY-K6303)– iCMs protocol.

1. Reagent Preparation: Prepare complete cardiomyocyte dissociation solution by mixing Cardiomyocyte Dissociation Reagent A and Cardiomyocyte Dissociation Reagent B at a 1:1 ratio.

Note: The prepared complete cardiomyocyte dissociation solution may be stored at −20°C.

2. Cell Dissociation: Remove the original culture medium and wash the cells once with DPBS. Add 500 μL of complete cardiomyocyte dissociation solution to each well and incubate at 37°C for 12–15 min, until the intercellular gaps enlarge and the cell bodies retract.

3. Termination of Dissociation: Centrifuge at 150× g for 3 min and discard the dissociation solution. Gently resuspend the cells in cardiomyocyte progenitor differentiation medium supplemented with CEPT Cocktail Plus.

4. Cell Seeding and Confocal Imaging Identification.

1. This product is sterile and should be handled using aseptic techniques. It is recommended to aliquot and store to avoid contamination.

  • Do most proteins show cross-species activity?

    Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
