MCE Cardiomyocyte Dissociation Solution is specifically designed for the dissociation of cardiomyocytes following PSC differentiation and selection. It contains highly active collagenase, dispase, and neutral protease, which work synergistically with a low concentration of recombinant serine protease to efficiently and gently dissociate densely adherent cardiomyocytes into a single-cell suspension.

Description & Advantages
MCE Cardiomyocyte Dissociation Solution is specifically designed for the dissociation of cardiomyocytes following PSC differentiation and selection. It contains highly active collagenase, dispase, and neutral protease, which work synergistically with a low concentration of recombinant serine protease to efficiently and gently dissociate densely adherent cardiomyocytes into a single-cell suspension. Following dissociation, digestion can be directly terminated using Cardiomyocytes (iCMs) Maturation Medium without the need to add a protease inhibitor. The resulting cardiomyocytes maintain high viability and functional activity, while preserving membrane potential integrity and physiological rhythmicity to the greatest extent possible.
This product is recommended for use in conjunction with the Human iPSC/ESC Cardiomyocyte Induction and Differentiation Kit (Cat. No. HY-K6303).
Protocol

1. Reagent Preparation: Thaw the cardiomyocyte dissociation solution overnight in a refrigerator at 4 °C prior to use.
Note: After the initial thawing, it is recommended to aliquot the solution into appropriate volumes for storage to avoid repeated freeze-thaw cycles.
2. Cell Dissociation: Remove the original culture medium and wash the cells three times with DPBS. Add 500 μL of cardiomyocyte dissociation solution to each well and incubate at 37°C for 12–15 min, until the intercellular gaps enlarge and the cell bodies retract.
3. Termination of Dissociation: Centrifuge at 150× g for 3 min and discard the dissociation solution. Gently resuspend the cells in cardiomyocyte progenitor differentiation medium supplemented with CEPT Cocktail Plus.
4. Cell Seeding and Confocal Imaging Identification.
