MCE Alcian Blue Staining Solution (pH 2.5) is commonly used for the detection of acidic mucopolysaccharides and mucins in histological studies. It is suitable for cartilage staining, visualization of mucin distribution, evaluation of intestinal metaplasia in gastrointestinal tissues, and the auxiliary identification of mucin-producing epithelial tumors.

Alcian Blue, also known as Alcian or Ingrain Blue, is a cationic dye with a copper phthalocyanine–like structure that is widely used in histological and pathological studies for the detection of acidic mucopolysaccharides and mucins. Its molecular structure consists of a central copper phthalocyanine ring linked to four isothiouronium groups through thioether bonds. These moderately basic isothiouronium groups confer a positive charge to the dye, enabling it to interact with negatively charged acidic groups in tissues and form stable insoluble complexes.

Under different pH conditions, Alcian Blue can selectively bind to different types of acidic groups and produce distinct staining reactions. When the staining system is adjusted to pH 2.5, carboxyl groups (—COO-) in tissues become ionized and form salt linkages with the positively charged dye, resulting in blue staining of carboxyl-containing acidic mucopolysaccharides and acidic mucins, such as hyaluronic acid and epithelial acidic mucins. In contrast, neutral mucins generally do not react with Alcian Blue and therefore remain unstained. By controlling the pH of the staining system, Alcian Blue staining can also be used to distinguish different types of acidic mucosubstances.

 

MCE Alcian Blue Staining Solution (pH 2.5) is commonly used for the detection of acidic mucopolysaccharides and mucins in histological studies. It is suitable for cartilage staining, visualization of mucin distribution, evaluation of intestinal metaplasia in gastrointestinal tissues, and the auxiliary identification of mucin-producing epithelial tumors.

This product is supplied with a Nuclear Fast Red counterstaining solution, which stains cell nuclei and enhances the contrast of tissue structures, thereby facilitating morphological observation and analysis.

Protocol

Reagents Preparation

1. Fixative: 10% formalin

2. Other reagents: xylene, neutral mounting medium or mounting reagent, graded ethanol solutions, distilled water, etc.

 

Procedure

1. Deparaffinization and Rehydration: Paraffin sections are first deparaffinized in xylene, followed by treatment with graded ethanol solutions, and then rehydrated in distilled water.

2. Acidification: Immerse the sections in Alcian Blue Acidification Solution for 3 min.

3. Alcian Blue Staining: Incubate the sections in Alcian Blue Staining Solution for 15-30 min, followed by rinsing under running water for 5 min.

4. Counterstaining: Counterstain the sections with Nuclear Fast Red Solution for 10 min, then rinse under running water for 1 min.

5. Dehydration, Clearing, and Mounting: Dehydrate the sections sequentially through graded ethanol solutions, clear with xylene, and finally mount with neutral mounting medium or mounting reagent.

 

Interpretation of Staining Results

1. Acidic mucins (e.g., sulfated mucins and sialomucins): Blue

2. Proteoglycans and hyaluronic acid: Blue.

3. Cell nuclei: Red

Attention

1. It is recommended to fix samples with 10% neutral buffered formalin to ensure optimal preservation of tissue morphology and structure.

2. For selective differentiation between sulfated mucins and proteoglycans, Alcian Blue staining at pH 1.0 (HY-K0603) can be used. Under this pH condition, the dye preferentially binds to sulfate groups. The staining procedure is generally the same as that for Alcian Blue (pH 2.5); however, the staining time may be appropriately extended to obtain optimal staining results.

3. Since ethanol is highly volatile, graded ethanol solutions should be freshly prepared before use to ensure consistent dehydration performance.

4. After opening, reagents are recommended to be used within 6 months. The bottle cap should be tightly closed after each use to prevent evaporation or deterioration.

5. After staining with this kit, subsequent immunofluorescence or other fluorescence-based staining is not recommended, as it may interfere with fluorescence signals or the interpretation of staining results.

  • Do most proteins show cross-species activity?

    Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
