TIE-1

TIE-1 is an endothelium-restricted receptor tyrosine kinase that combines immunoglobulin-like and EGF-like ectodomains with three fibronectin type III repeats. Unlike its family partner TIE-2, no unequivocal activating ligand has been established, and TIE-1 is generally considered an orphan or modulatory receptor. It forms heterodimers with TIE-2 and fine-tunes angiopoietin-driven signaling, and mouse genetics shows TIE-1 is required for vascular maturation and vessel integrity during development and quiescence. A circulating cleaved ectodomain, TIE1ECD, rises in sepsis and preeclampsia, supporting its use as an endothelial activation biomarker in critical care. These features position TIE-1 as both a modulator of angiopoietin signaling and a source of clinically informative circulating fragments.
