Description & Advantages
Firefly Luciferase (Fluc) mRNA (N1-methyl-pseudouridine-modified) is an in vitro transcribed (IVT) mRNA encoding Firefly Luciferase (Fluc), in which uridine triphosphate (UTP) is completely or partially replaced with N1-methyl-pseudouridine triphosphate (N1-Me-pUTP, also referred to as m1ΨTP). Incorporation of N1-methyl-pseudouridine reduces the innate immunogenicity of mRNA while enhancing its stability and protein expression.

Protocol
1. Before opening the vial, briefly centrifuge at 2,000–4,000 g for 3 min to collect the lyophilized mRNA at the bottom of the tube.
2. Reconstitute the mRNA with Nuclease-Free Water to prepare a stock solution at a concentration of ≥1 μg/μL. Aliquot the stock solution according to the amount required for each experiment.
Note: a. The reconstituted mRNA stock solution may be stored at −80°C for up to 6 months. Repeated freeze-thaw cycles should be avoided to preserve mRNA integrity and biological activity.
b. Throughout the procedure, use only RNase-free reagents, consumables, and laboratory equipment to prevent RNase contamination and subsequent mRNA degradation.
