Description & Advantages

Lentivirus vector is a gene therapy vector based on HIV-1, which can stably integrate exogenous genes into the host cell genome. It offers the advantages such as an extensive infection spectrum, effective infection and division period, static cell cells, long-term stable expression of exogenous genes, etc. It has become a powerful tool for importing exogenous genes, and has been widely used in gene overexpression in various cell lines, RNA interference, microRNA research and in vivo animal experiments.

MCE Lentivirus Transfection Reagent is a new type of transfection reagent based on cationic polymer, which is suitable for lentivirus packaging and transfection. It significantly improves the efficiency of packaging and transfection, resulting in the production of more recombinant lentivirus. The viral titer of the control plasmid obtained by transfection and packaging can reach 108 TU/mL, which has the advantages of high infection efficiency, wide infection spectrum, high reproducibility and simple operation.

Protocol

1. Preparation of Packaging Cells

Seed HEK293T or other lentiviral packaging cells before transfection so that cell density is approximately 70%-80% at transfection. Use healthy, actively growing cells.

2. Preparation of Transfection Complexes

GFP Control Plasmid 4 μg
Helper Mix 4 μL
Lentivirus Transfection Reagent 40 μL

1. Dilute the transfer plasmid and Helper Mix in a suitable serum-free medium and mix.

2. Dilute Lentivirus Transfection Reagent separately in serum-free medium.

3. Add the diluted transfection reagent to the plasmid mixture, mix gently, and incubate for approximately 15 min at room temperature.

3. Lentivirus Packaging

1. Add the transfection complexes evenly to the packaging-cell culture and continue normal culture.

2. Collect the first viral supernatant approximately 48 h after transfection and add fresh medium.

3. Collect viral supernatant again at approximately 72 h and pool the two harvests.

4. Filter, concentrate, determine viral titer, or use the virus for downstream infection as required.

Note: Lentivirus packaging, harvest, infection, and waste disposal should be performed under appropriate BSL-2 biosafety conditions.
