This product is serum-free and supports cell growth and survival. It has been associated with the growth and expression of neuroblastoma, and with post-mitotic neuronal survival and expression in primary cultures of both the peripheral nervous system (PNS) and central nervous system (CNS). This product is formulated with water-for-injection. The 5 mL is defined as the base specification. All larger sizes correspond to incremental volumes of this base.

Description & Advantages

This product is serum-free and supports cell growth and survival. It has been implicated in enhancing the growth and expression profiles of neuroblastoma cells, as well as in promoting the survival and phenotypic expression of post-mitotic neurons in primary cultures derived from both the peripheral nervous system (PNS) and central nervous system (CNS). It is widely used for the cultivation of neural cell lines and for the culture and directed differentiation of pluripotent stem cells. Additionally, it is compatible with growth factors such as bFGF and EGF when incorporated into serum-free neuronal culture media based on DMEM or DMEM/F12.

MCE N-2 Supplement (100×), Sterile is a chemically-defined, serum-free supplement based on Bottenstein’s N-1 formulation, the solution contains transferrin, recombinant insulin, luteinizing hormone, putrescine, and selenite.

Protocol

Cell Culture Supplementation

1. Completely thaw N-2 Supplement (100×), Sterile and mix gently before use.

2. Under sterile conditions, add the supplement to sterile cell culture medium at 1 : 100 (v/v) to obtain a final working concentration of 1×.

3. Mix thoroughly and use directly for the appropriate cell culture.

Note: The solution may show color differences after freezing but can fully dissolve in a 37°C water bath. Aliquoting is recommended to avoid repeated freeze-thaw cycles.
