MCE One Step Colored Rapid Gel Preparation Kit (12.5%) uses pre-mixed formulations of the upper gel and lower gel. Simply mix the reagents in pairs, add modified coagulation agent, and the gel will form quickly and easily.

Description & Advantages
MCE One Step Colored Rapid Gel Preparation Kit (12.5%) uses pre-mixed formulations of the upper gel and lower gel. Simply mix the reagents in pairs, add modified coagulation agent, and the gel will form quickly and easily. The prepared upper gel is colored, making sample loading and distinguishing different gels convenient. The kit comes with a modified polymerizing agent, APS, which has excellent stability and coagulating effects. No need to add TEMED during gel preparation.
The 125-gel version can prepare 125 mini PAGE gels (0.75 mm thickness). The prepared gels can be used for both denaturing and non-denaturing PAGE gel electrophoresis.
Protocol
Take the preparation of one mini gel with a thickness of 0.75/1.0/1.5 mm as an example.

Procedure
1. Invert and mix each component 6-8 times before use.
2. According to the required gel thickness, mix equal volumes of One-Step Lower Gel Solution (2×) and One-Step Lower Gel Buffer (2×). Add the corresponding volume of Modified Gel Accelerator and mix thoroughly.
3. Immediately transfer the prepared lower gel solution into the glass plate. The distance between the liquid surface and the top edge of the short glass plate should be 0.5 cm longer than the comb tooth length.
Note: 1) The prepared solution volume is excessive; do not load the entire volume. Retain a small amount of solution in the tube to monitor the gelation status.
2) Avoid vigorous shaking during mixing. Prevent air bubbles from being introduced into the gel casting glass plate during pouring.
3) When the room temperature is low, the gelation rate may decrease; appropriately extend the gelation time.
4. According to the required gel thickness, mix equal volumes of One-Step Upper Gel Solution (2×) and One-Step Colored Upper Gel Buffer (2×). Add the corresponding volume of Modified Gel Accelerator and mix thoroughly.
5. Immediately transfer the prepared upper gel solution into the glass plate, insert the comb, and remove the comb after the upper gel is polymerized (15 min). The gel is then ready for electrophoresis.
Note: 1) Freshly prepared electrophoresis buffer is recommended.
2) Recommended electrophoresis conditions: 150 V for approximately 55 min or 200 V for approximately 40 min.
Storage
One Step Colored Upper Gel Buffer (2×), One Step Upper Gel Solution (2×), One Step Lower Gel Solution (2×), One Step Lower Gel Buffer (2×): 4°C, 1 year.
One Step Modified Coagulation Agent: -20°C, 1 year. Avoid repeated freeze-thaw cycles.
Attention
1. The upper gel prepared by this product does not concentrate samples, similar to pre-cast gels. Compared to traditional PAGE gels, it provides better protein band separation, with small proteins (e.g., 10 kDa) being clearly resolved, and the protein bands are narrower and sharper.
2. The amount of modified polymerizing agent provided is for reference. The actual amount can be adjusted based on personal habits and experience. Adding more polymerizing agent speeds up the gel formation, and less slows it down. Under the same conditions, the higher the temperature, the faster the gel forms. If the temperature is too high, consider reducing the amount of polymerizing agent.
3. Allow the gel solutions and buffers to return to room temperature before use to effectively prevent bubble formation during gel preparation.
4. After opening, the modified polymerizing agent can be stored at 4°C for at least 3 months. For long-term storage, it is recommended to aliquot and store at -20°C.
