Description & Advantages
RNA interference (RNAi) refers to the small double-stranded RNAs that can specifically degrade homologous mRNAs, inhibiting or shutting down the expression of the corresponding genes. By utilizing RNAi technology to design small interfering RNA (siRNA) targeting the mRNA of disease-causing genes, it is possible to specifically knock out or turn off the expression of specific genes.
MCE siRNA/miRNA Transfection Reagent is a novel cationic polymer transfection reagent for siRNA and miRNA transfection. The exogenous siRNA/miRNA forms a stable complex with the transfection reagent and enters the cell through cytosis, subsequently releasing the siRNA/miRNA in the cytoplasm to achieve cellular transfection of siRNA/miRNA.

Protocol
1. Cell Preparation
Seed cells before transfection so that they are healthy and actively growing. In MCE validation, approximately 40% cell density produced good siRNA transfection performance.
2. Preparation of Transfection Complexes

1. Dilute siRNA/miRNA and siRNA/miRNA Transfection Reagent separately in serum-free medium.
2. Add the diluted transfection reagent to the diluted siRNA/miRNA, mix gently, and incubate for approximately 15 min at room temperature.
3. Add the complexes to the cell culture and mix gently. Serum in complete medium generally does not significantly affect transfection, and medium replacement is not mandatory solely for the transfection step.
3. Concentration Optimization and Analysis
siRNA or miRNA mimic may be optimized from approximately 10-60 pmol/well or approximately 10-200 nM. miRNA inhibitors generally require a higher starting concentration. In MCE validation, 1 μL siRNA (20 μM) with 2 μL Transfection Reagent produced efficient gene silencing.
Perform downstream mRNA, protein-expression, or fluorescence analysis approximately 24-48 h after transfection.
