Etanercept, a dimeric fusion protein that binds TNF, acts as a TNF inhibitor. Etanercept competitively inhibits the binding of both TNF-α and TNF-β to cell surface TNF receptors, rendering TNF biologically inactive. Etanercept shows efficacy against rheumatoid arthritis, juvenile idiopathic arthritis, and plaque psoriasis.

Efruxifermin is an Fc-FGF21 fusion protein (human IgG1 Fc domain linked to a modified human FGF21). Efruxifermin has prolonged half-life and enhanced receptor affinity compared with native human FGF21. Efruxifermin can be used for the research of non-alcoholic steatohepatitis.

Abatacept (CTLA4lg) is a soluble fusion protein consisting of the extra-cellular domain of human CTLA4 and a fragment of the Fc portion of human IgG1 (hinge and CH2 and 3 domains). Abatacept is a selective T-cell co-stimulation modulator and a protein agent for the autoimmune diseases.

Belatacept (BMS 224818) is a selective T-cell costimulation blocker and a costimulator of the CD28-CD80/86 pathway. Belatacept binds to the CD 80/86 ligand and inhibits CD-28-mediated T cell costimulation and IFN-γ production. Belatacept can be used in studies of immunosuppression in organ transplantation. Sotatercept (ACE-011) is a soluble activin receptor 2A (ACVR2A) type IgG Fc fusion protein. Sotatercept combines activin and growth differentiation factor to try to restore the balance between growth promotion and growth inhibition signal pathways. Sotatercept has potential application in pulmonary arterial hypertension, anemia, bone loss, erythropoiesis, multiple myeloma (MM) osteolytic lesions.

Luspatercept (ACE-536) is a recombinant modified ActRIIB fusion protein that binds with transforming growth factor β superfamily ligands. Luspatercept increases the erythrocyte numbers and promotes maturation of erythroid precursors. Luspatercept binds with GDF11 and inhibits Smad2/3 signaling. Luspatercept can be used for the research of anemia.

Aritinercept (AUR-200) is a recombinant fusion protein targeting TNFSF13B/BAFF/CD257. Aritinercept neutralizes BAFF and APRIL (TNFSF13), reduces B cell proliferation, suppresses peripheral B cells, lowers serum immunoglobulins. Aritinercept reduces renal damage markers, decreases IFNγ, IL-17A, and anti-dsDNA autoantibodies, and improves survival in a mouse model of SLE. ritinercept can be used for the research of systemic lupus erythematosus.

Eftezirleukin alfa is a human immunoglobulin G4 (IgG4) Fc fragment. Eftezirleukin alfa is associated with IL-2.

Telitacicept (RC18) is a fully human TACI-Fc fusion protein. Telitacicept is a dual B lymphocyte stimulator (BLyS)/APRIL (a proliferation-inducing ligand) inhibitor that effectively blocks proliferation of B lymphocytes. Povetacicept (ALPN-303) is an engineered Fc fusion protein containing the TACI domain, and acts as a dual APRIL/BAFF antagonist. The K_d value of Povetacicept for human BAFF is 59.3 pM, while its K_d value for human APRIL is 1.00 pM. Povetacicept reduces the activation, proliferation, differentiation and survival of B cells, and inhibits the production of immunoglobulins and autoantibodies. Sotatercept (mIgG2a) mainly inhibits the SMAD2/3 signaling pathway, and can be used in various diseases such as chronic kidney disease. Pabinafusp alfa (JR-141) is a transferrin receptor-targeting antibody consisting of Iduronate 2-sulfatase (HY-P76399) and an anti-human transferrin receptor antibody. Pabinafusp alfa is blood-brain permeable and prevents heparan sulfate (HS) deposition in the central nervous system of mucopolysaccharidosis II (MPS II) mice. Pabinafusp alfa improves learning and prevents central nervous system neuronal damage in mice.

Atrosimab is an Fv-Fc1K fusion protein with an EC₅₀ value of 0.37 nM against humans. Atrosimab inhibits TNF-induced TNFR1 activation, release of IL-6 and IL-8, and cell death, and alleviates neuroinflammation. Dazodalibep (MEDI 4920; VIB 4920) is a non-antibody fusion protein that targets CD40L. IBI-363 is a PD-1/IL-2 bispecific antibody with functions of blocking the PD-1/PD-L1 pathway and activating the IL-2 pathway. The IL-2 arm of IBI-363 retains affinity for IL-2Rα but attenuates binding ability to IL-2Rβ and IL-2Rγ to reduce toxicity. The PD-1 binding arm of IBI-363 enables PD-1 blockade and selective delivery of IL-2. IBI-363 can be used in cancer research, such as non-small cell lung cancer.

Efineptakin alfa (NT-17) is a long-acting recombinant human IL-7. Efineptakin alfa supports the proliferation and survival CD4⁺ and CD8⁺ cells in both human and mice. Efineptakin alfa can be used for glioblastoma research.
