Description & Advantages
MCE CEPT Cocktail Plus (1000×) is a composite supplement specifically formulated for pluripotent stem cell culture. It contains three key small molecules-Chroman 1 (antioxidant), Emricasan (caspase inhibitor), and Trans-ISRIB (integrated stress response modulator)—and is further supplemented with Polyamine Solution, L-ascorbic acid phosphate (LAA), niacin, cysteine, and other auxiliary components.
Through synergistic actions that suppress oxidative damage, block apoptotic signaling, and regulate protein translation, MCE CEPT Cocktail Plus (1000×) effectively reduces cellular stress via multiple pathways, resulting in markedly improved viability and cloning efficiency of pluripotent stem cells. In addition, CEPT Cocktail Plus (1000×) enhances culture stability during routine passaging, promotes post-thaw recovery following ultra-low-temperature cryopreservation, and supports efficient formation of embryoid bodies and diverse organoids (such as intestinal and brain organoids).
Protocol
1. Thaw CEPT Cocktail Plus (1000×) at 4°C in advance.
Note: a. Aliquot as needed to minimize repeated freeze/thaw cycles.
b. Due to localized concentrations of polyamines, a milky or flocculent precipitate may appear during storage at 4°C. Mix thoroughly before use.
2. Add CEPT Cocktail Plus (1000×) to the culture medium at a 1:1000 ratio. Example: Add 10 μL of CEPT Cocktail Plus (1000×) to 10 mL of culture medium.
3. Filter the prepared complete medium through a 0.22 μm membrane before use.
Note: It is recommended to use the medium immediately after preparation; long-term storage is not recommended.
