Description & Advantages
MCE Exosome Protein Lysis Buffer is a lysis buffer specifically designed for exosome samples, capable of efficiently lysing exosome proteins. The lysed protein samples can be used in experiments such as BCA, SDS-PAGE, and Western Blot.
This product contains various inhibitors including sodium deoxycholate, EDTA, PMSF, and a Protease Inhibitor Cocktail (AEBSF, Aprotinin, Bestatin, E-64, Leupeptin, Pepstatin A), which effectively prevent protein degradation.
Protocol
1. Experimental Preparation
Before the experiment, thaw the Exosome Protein Lysis Buffer at 4°C. Once thawed, take an appropriate amount and keep it on ice for use.
Note: It is recommended to aliquot the lysis buffer after the first thaw to avoid repeated freeze-thaw cycles.
2. Exosome Lysis
Mix the exosome sample with the lysis buffer at a 1:1 volume ratio, and incubate on ice for 10 min.
3. Protein Collection
Centrifuge at 4°C, 11,200 g (12,000 rpm) for 5 min, and collect the supernatant. The protein concentration in the supernatant can be quantified using the BCA method, and then used for Western Blot and other subsequent experiments.
Note: a. The above centrifugation is based on a small centrifuge (effective radius ~ 7 cm) and ≤ 2 mL centrifuge tubes. For larger volumes, it is recommended to centrifuge in aliquots.
b. The protein concentration after lysis should be controlled below 2 mg/mL to ensure complete lysis.

Attention
1. All steps of protein extraction must be performed on ice or at 4°C to maintain protein activity.
2. Due to the high detergent concentration, the Bradford assays cannot be used to measure protein concentration in samples obtained using this lysis buffer.
