Description & Advantages
Basement Membrane Matrix is a natural basement membrane preparation extracted from a mouse tumor rich in extracellular matrix (ECM) proteins. It mimics the in vivo cellular microenvironment and provides structural support for cell growth. The matrix contains abundant ECM components, including: Laminin, Collagen IV, Entactin/Nidogen, Heparan Sulfate Proteoglycans (HSPGs). It also contains various naturally occurring growth factors, including: Insulin-like Growth Factor-1 (IGF-1), Transforming Growth Factor-β (TGF-β), Vascular Endothelial Growth Factor (VEGF), Epidermal Growth Factor (EGF), Basic Fibroblast Growth Factor (bFGF).

The convenient ready-to-use format allows immediate use after opening, minimizing hands-on preparation, streamlining workflow, improving experimental efficiency, and ensuring excellent batch-to-batch consistency and reproducibility.
Protocol
Remove the matrix gel and place it on crushed ice for use.
Add an appropriate amount of matrix gel to the center of the culture plate or dish to completely cover the growth surface. Add the gel slowly and gently tilt or swirl the vessel to distribute it evenly and avoid bubble formation. Recommended volumes for different culture vessels are listed below.

Incubate the coated culture vessel at 37°C for at least 60 min until the matrix gel is fully gelled. If not used immediately, seal the vessel with sealing film, store at 4°C, and use within one week.
Before use, allow the coated culture vessel to equilibrate at room temperature for 1 h. Carefully aspirate the supernatant, then slowly add room-temperature human pluripotent stem cell (PSC) culture medium (MCE Cat. No.: HY-K6501) containing CEPT Cocktail Plus (1000×) (MCE Cat. No.: HY-K6021) along the wall of the vessel.
Note: Avoid touching or scratching the matrix-coated surface with the pipette tip during operation, as this may affect the coating performance.
Seed the stem cells into the coated culture vessel.
Continue incubation in a cell culture incubator. After 18-24 h, examine cell attachment. If the cells are well attached and show normal morphology, replace the medium with human pluripotent stem cell (PSC) culture medium without CEPT Cocktail Plus and continue culture.
Replace the culture medium daily thereafter. When cell confluency reaches 50% or higher, the medium volume may be increased appropriately according to cell growth to support continued proliferation.

Attention
1. This product has been optimized for ready-to-use applications. Further dilution is not recommended, as it may affect coating performance.
2. Keep the product on crushed ice throughout the entire procedure. Maintain low-temperature conditions at all times to prevent premature gelation of the matrix.
3. Pre-cool all consumables (pipette tips, tubes, culture plates, etc.) before use.
4. Avoid holding the lower portion of containers containing matrix to prevent localized warming and gelation.
5. This product is for R&D use only, not for drug, household, or other uses.
6. For your safety and health, please wear a lab coat and disposable gloves to operate.
