Description & Advantages

MCE Stem Cell Cryopreservation Medium is a ready-to-use, serum-free Cryopreservation Medium optimized for human pluripotent stem cells (PSCs). By precisely balancing DMSO concentration and osmolarity, the formulation enhances post-thaw viability and preserves PSC functional integrity. It contains a composite low-toxicity cryoprotectant system together with the CEPT cocktail, which minimizes ice crystal formation, oxidative stress, and osmotic damage while maintaining mitochondrial function and cytoskeletal stability. Stabilizing serum albumin is included to improve membrane protection and increase cellular tolerance during freezing.

PSCs preserved with this medium exhibit high post-thaw viability (> 90%) and stable expression of pluripotency markers such as OCT4 and NANOG, providing consistent and reliable cell sources for subsequent expansion and differentiation. This product is suitable for long-term, stable storage of PSCs and downstream applications.

Protocol

1. Cell Harvesting

When cells reach approximately 85% confluency, they are ready for cryopreservation. For a 6-well plate, each well typically yields enough cells for about 2 cryovials.

2. Preparation of Cryovials

Pre-label the required number of 1.5/2 mL cryovials with the cell name, passage number (P#), and date.

3. Pre-treatment of Cryopreservation Medium

Remove the stem cell cryopreservation medium from -20°C and thaw at 4°C. Before use, bring it to room temperature and mix thoroughly.

4. Cell Washing

Aspirate the culture medium and add 2 mL of DPBS (calcium- and magnesium-free, Cat. No.: HY-K3008) to each well. Gently mix to wash the cells, and then aspirate the wash solution.

5. Cell Dissociation

Add 2 mL of stem cell dissociation solution (Cat. No.: HY-K6020) to each well and incubate the plate in a 37°C incubator for 5-7 min。

6. Termination of Dissociation

Remove the plate and aspirate the dissociation solution.

7. Addition of Cryopreservation Medium and Aliquoting

Thoroughly mix the stem cell cryopreservation medium brought to room temperature. Add 1 mL per well, gently pipette up and down 3 times, and mix by gently rocking in a horizontal “cross” motion. Transfer 500 μL of the cell suspension into each 1.5/2 mL cryovial.

8. Controlled Freezing and Storage

Place the cryovials into a controlled-rate freezing container and store at -80°C overnight. The next day, transfer the cryovials into a liquid nitrogen tank for long-term storage.
