ADAM10

ADAM10, the mammalian ortholog of Drosophila Kuzbanian, is the principal constitutive alpha-secretase of the ADAM family. It performs the non-amyloidogenic cleavage of amyloid precursor protein (APP) and the S2 cleavage of Notch receptors, and also sheds ligands such as Delta, Jagged and fractalkine together with surface molecules like CD23, L-selectin and VE-cadherin. This breadth makes ADAM10 central to neurodevelopment, Notch and EGFR signaling, and endothelial biology. Selective inhibitors and activators of ADAM10 are actively explored for Alzheimer disease, inflammation and cancer. In the immune system, ADAM10 is required for Notch signaling in T cells, and its shedding of VE-cadherin has been linked to vascular permeability.
