Hanks’ Balanced Salt Solution (HBSS) is widely used in various cell culture applications, including washing cells prior to dissociation, transporting cells or tissue samples, diluting cells for cell counting, and preparing a variety of reagent solutions. Formulations without calcium and magnesium are suitable for washing chelating agents prior to cell dissociation procedures. The 500 mL is defined as the base specification. All larger sizes correspond to incremental volumes of this base.

Description & Advantages
Balanced Salt Solution (BSS), also known as Physiological Solution, combines the buffering capacity of a buffer system, the isotonic properties of physiological saline, and the basic nutrient supply characteristics of culture media. It functions to maintain osmotic balance, stabilize pH, and provide essential nutrients, thereby supporting the basic survival and metabolic requirements of tissues, organs, or cells in vitro. Some balanced salt solutions also contain a small amount of phenol red as a pH indicator.

Protocol
Direct Use
1. HBSS, Phenol Red, Calcium, Magnesium is a ready-to-use sterile 1× balanced salt solution and does not require dilution.
2. Equilibrate to room temperature or the required temperature before use.
3. Use directly for cell washing, short-term cell/tissue incubation, sample dilution, and preparation of related experimental reagents.
4. Remove the required volume under aseptic conditions, avoid contaminating the original bottle, and close the container promptly after use.
