MCE 3-Color Prestained Protein Marker (10-180 kDa) consists of 10 prestained recombinant proteins with an apparent molecular weight range of 10–180 kDa. This marker enables real-time monitoring of protein migration during SDS-PAGE and facilitates evaluation of protein transfer efficiency. The 250 μL is defined as the base specification. All larger sizes correspond to incremental volumes of this base.

Description & Advantages
MCE 3-Color Prestained Protein Marker (10-180 kDa) consists of 10 prestained recombinant proteins with an apparent molecular weight range of 10–180 kDa. In the Tris-Glycine electrophoresis system, the 75 kDa band is orange, the 25 kDa band is green, and the remaining bands are blue (10, 17, 33, 43, 55, 95, 130, and 180 kDa).
This marker enables real-time monitoring of protein migration during SDS-PAGE and facilitates evaluation of protein transfer efficiency. Following electrophoresis, the prestained proteins can be efficiently transferred onto PVDF or nitrocellulose (NC) membranes while retaining distinct colored bands for convenient molecular weight estimation and assessment of transfer performance.
This product is supplied in a ready-to-use format and can be loaded directly without further preparation. Do not heat before use. The storage buffer contains 25 mM Tris-H3PO4 (pH 7.5), 1 mM EDTA, 2% (w/v) SDS, 10 mM DTT, 1 mM NaN3, and 33% (v/v) glycerol.
Protocol
1. Before use, thaw the Prestained Protein Marker completely at room temperature. Gently mix after thawing. Do not heat.
2. Load 5 μL of the marker and perform SDS-PAGE electrophoresis together with the experimental samples. For first-time use, the loading volume may be optimized according to preliminary experimental results and specific application requirements.

Attention
1. Do not heat, dilute, or add reducing agents to this product.
2. It is recommended to aliquot the marker into single-use portions to avoid repeated freeze-thaw cycles.
3. When used with electrophoresis systems other than the Tris-Glycine system or with different gel types, the apparent molecular weights of the prestained protein bands may vary. Please refer to the Appendix of this manual for the corresponding molecular weight information. Because the prestaining dyes carry electrical charges, they affect protein migration in different electrophoresis buffer systems, resulting in differences in the apparent molecular weights.
4. For Western blot transfer of high-molecular-weight proteins, extending the transfer time or increasing the transfer voltage is recommended to achieve optimal transfer efficiency.
5. When using low-percentage polyacrylamide gels, some low-molecular-weight protein bands may migrate ahead of the bromophenol blue dye front. This is a normal phenomenon.
