Description & Advantages
DMEM/F-12 (Dulbecco’s Modified Eagle Medium/Nutrient Mixture F-12) is a widely used basal medium suitable for the culture of a variety of mammalian cells, including MDCK cells, glial cells, fibroblasts, and human endothelial cells.
MCE Advanced DMEM/F-12 is an optimized mammalian cell culture medium designed to reduce the requirement for fetal bovine serum (FBS). Compared with classical DMEM/F-12 medium, it can reduce serum usage by 50–90% while maintaining comparable cell growth rates and cellular morphology, thereby effectively lowering serum consumption and reducing overall cell culture costs.
This medium is suitable for a wide range of cell lines, many of which can be cultured directly without adaptation. Cell lines that have been successfully cultured without adaptation include Vero, LX-2, KGN, C8-D1A, HeLa, KLE, ARPE-19, NIH-3T3, 4T1, JHH-7, and NCI-H2009.
MCE offers various formulations of DMEM medium to meet the diverse requirements of different cell culture applications.
Protocol
Add thawed serum, antibiotics, and other required supplements, such as L-glutamine or L-alanyl-L-glutamine (Cat. No.: HY-K1046/HY-K1046P), to the medium at the desired ratios. Mix thoroughly to prepare complete culture medium.
Note: a. A final fetal bovine serum (FBS) concentration of 1–5% is recommended. Optimization of FBS concentration for individual cell lines is recommended to minimize serum usage whenever possible.
b. DMEM employs a sodium bicarbonate buffering system, it requires a 5–10% CO2 atmosphere to maintain physiological pH stability.
