Description & Advantages
Basement Membrane Matrix is a natural basement membrane extract derived from mouse tumor tissue rich in extracellular matrix (ECM) proteins. It mimics the in vivo cellular microenvironment and provides structural support for cell growth. It contains abundant extracellular matrix components, including laminin, type IV collagen (Collagen IV), entactin, heparan sulfate proteoglycans (HSPGs), and others. It also contains a variety of naturally occurring growth factors, including insulin-like growth factor-1 (IGF-1), transforming growth factor-β (TGF-β), vascular endothelial growth factor (VEGF), epidermal growth factor (EGF), basic fibroblast growth factor (bFGF), and others.
MCE Stem Cell Basement Membrane Matrix is specifically designed for stem cell culture applications. It promotes stem cell attachment and proliferation while helping maintain stemness. It is suitable for the culture, expansion, and differentiation of human embryonic stem cells (hESCs) and induced pluripotent stem cells (iPSCs), as well as the establishment and culture of stem cell-derived organoids. The phenol red-free formulation effectively avoids potential interference from phenol red and is suitable for experiments sensitive to background signals or requiring minimized color interference.
Protocol
MCE Stem Cell Basement Membrane Matrix has the following four coating methods.
| Coating Method | Primary Function | Common Applications |
|---|---|---|
| Thin Gel | Forms an approximately 0.5 mm gel layer | Primary cell culture, cell migration and invasion assays |
| Thin Coating | Does not form a gel; forms an ECM coating layer only | Primary cell culture, iPSC expansion, and cell adhesion assays |
| Thick Gel | Forms a 1-2 mm three-dimensional gel structure | Angiogenesis assays and aortic ring assays |
| Sandwich Coating | Cells are cultured between two layers of basement membrane matrix | Stem cell culture and differentiation, polarization, and morphogenesis studies |
Preparation Before Experiment
a. Slight variations in protein concentration may occur between different batches. Please refer to the corresponding Certificate of Analysis (COA) for specific information.
b. To achieve optimal experimental reproducibility, it is recommended to calculate the required volume based on the actual protein concentration and dilute the matrix using pre-chilled serum-free medium.
c. Maintain low temperature conditions (ice bath or 4°C) throughout the entire procedure to prevent premature gelation of the basement membrane matrix.
