Description & Advantages
MCE CEPT Cocktail is composed of CET small molecules (Chroman 1, Emricasan and Trans-ISRIB) and Polyamine Solution. This product enhances cell survival and promotes clonal growth of genetically stable hPSCs. In addition, this product provides strong improvements for several key applications in stem cell research, including routine cell passaging, cryopreservation of pluripotent and differentiated cells, embryoid body (EB) and organoid formation, etc.
Protocol
1. Preparation of stock solutions

2. Preparation of CEPT-containing culture medium
Add Chroman 1, Emricasan, and Trans-ISRIB stocks to complete hPSC culture medium at 1 : 10,000 immediately before use. For example, add 1 μL of each 10,000× stock to 10 mL complete medium.
3. Add Polyamine Solution at 1 : 1,000. For example, add 10 μL Polyamine Solution (1,000×) to 10 mL complete medium.
4. Mix thoroughly and sterile-filter through a 0.22 μm filter. The final CEPT medium contains 50 nM Chroman 1, 5 μM Emricasan, 0.7 μM Trans-ISRIB, and 1× Polyamine.
Note: CEPT Cocktail can be used for routine hPSC passaging, single-cell cloning, cryopreservation/recovery, EB formation, organoid formation, and related applications. Use all four components together for optimal cytoprotection.
