Description & Advantages
Apoptosis is a form of programmed cell death. Based on the cellular changes during programmed cell death, apoptosis is often classified into early, middle, and later stages. The later stages of apoptosis are characterized by changes in nuclear morphology, chromatin condensation, nuclear envelope degradation, and DNA fragmentation.
YO-PRO-1 (YP1), also known as Oxazole Yellow, is a non-cell-membrane-penetrating green fluorescent dye. It does not fluoresce without binding to DNA, but emits green fluorescence once bound to DNA. Propidium Iodide (PI) is a cell-membrane impermeable dye that can stain late-stage apoptosis cells and necrosis cells with red fluorescence.
MCE YO-PRO-1/PI Apoptosis and Necrosis Detection Kit is a dual-fluorescence method based on the green fluorescent dye YO-PRO-1 (YP1) and the red fluorescent dye Propidium Iodide (PI) for detecting cell apoptosis and necrosis. After co-staining with YO-PRO-1 and PI, normal cells are essentially non-fluorescent, apoptosis cells show green fluorescence, and necrosis cells show red and green fluorescence.
This kit is compatible with various fluorescence detection systems, including fluorescence microscopy, flow cytometry, fluorescence enzyme labeling, and other fluorescence detection systems.
Protocol
1. Culture and treat cells according to the experimental design and collect approximately 1 × 105-1 × 106 cells.
2. Wash cells once with PBS or a suitable serum-free buffer and resuspend in 1 mL buffer.
3. Add 1 μL YO-PRO-1 Staining Solution and 1 μL PI Staining Solution per 1 mL cell suspension and mix gently.
4. Incubate for approximately 20-30 min at room temperature protected from light.
5. Analyze by fluorescence microscopy, flow cytometry, or fluorescence microplate reader.
Interpretation
Normal viable cells show little or no fluorescence; apoptotic cells show mainly green YO-PRO-1 fluorescence; necrotic cells or cells with severely compromised membranes show both green YO-PRO-1 and red PI fluorescence.
