    Immunofluorescence (IF) analysis of SD rat cortical neurons: After 7 d of in vitro culture (10× magnification), three culture conditions were compared: no Bi-27 supplement (W/O), Bi-27 Serum-free Supplement (50×), Insulin-Free (HY-K3014C), and Gibco B-27 Supplement, minus insulin (Competitor). The results demonstrated that supplementation with Bi-27 significantly enhanced neuronal attachment, increased cell density, and promoted more extensive and well-developed neurite outgrowth. In contrast, the W/O group showed poor overall cell condition, characterized by reduced adhesion, decreased cell density, and limited neurite development. Further comparison indicated that HY-K3014C and the Gibco B-27 Supplement, minus insulin (Competitor), exhibited comparable performance in supporting the growth and differentiation of SD rat cortical neurons, with no statistically significant differences observed between the two groups. Overall, HY-K3014C demonstrated equivalent performance to the imported competitor in maintaining neuronal morphology, promoting cell growth, and supporting neurite development.

    Analysis of CTG assay and microscopic observation in colon cancer cells: After 6 days of in vitro culture, three culture groups were compared: no Bi-27 supplement (W/O), Bi-27 Serum-free Supplement (50×), Insulin-Free (HY-K3014C), and Gibco B-27 Supplement, minus insulin (Competitor). Cell growth status was comprehensively evaluated by microscopic observation (4×, 10×, 20× magnification) and CellTiter-Glo (CTG) luminescence detection. The results showed that compared with the Bi-27-free control group, supplementation with insulin-free B-27 improved the overall cellular growth, featuring more intact cell structure, clearer morphology of organoids/cell clusters and a more obvious increase in volume. Quantitative CTG results further revealed no statistically significant differences in cell viability among all groups (ns). Specifically, HY-K3014C and Gibco B-27 Supplement, minus insulin (Competitor) exerted equivalent effects on cell viability, with neither showing a significant difference relative to the control group. In conclusion, in this colon cancer cell culture system, HY-K3014C Bi-27 performs comparably to the competitor in supporting cell growth and preserving cell viability, with no remarkable functional differences detected.

As a neuronal cell culture supplement, it is specifically designed to support the growth and maintenance of hippocampal neurons and other central nervous system (CNS) neurons in applications where the presence of insulin may interfere with experimental outcomes.

    This product is an optimized serum-free supplement formulation enriched with multiple antioxidants to help reduce oxidative stress-induced cellular damage, thereby improving neuronal viability and culture stability.

    The recommended working concentration of Bi-27 Supplement is 2% (v/v), corresponding to a 1:50 dilution before use in the culture medium.

    Protocol

    Prepare complete Neuronal basal Medium

    1. Thaw MCE Bi-27 Serum-free Supplement (50×), Insulin-Free overnight at 4°C.

    2. Aseptically add 2% Bi-27 and 0.5mM L-Glutamine to the Neuronal Basal Medium before use.

    3. Primary Hippocampal Neuron Culture: Add 25 µM of L-glutamate to the complete Neuronal Basal Medium. The medium can be replaced after 4 days of culture, and glutamate should not be added to the medium after this change.

    Note: Once prepared, the complete Neuronal Basal Medium can be stored at 4°C for two weeks, protected from light.

    Attention

    1. This product appears as a light yellow liquid. The color may not be consistent following freezing, but it can fully dissolve in a 37°C water bath.

    2. It is recommended to appropriately aliquot the solution to avoid repeat freeze-thaw cycles.

    3. Pay attention to aseptic operation to avoid contamination.
