Description & Advantages
MCE Versene Solution is a gentle, enzyme-free, ready-to-use cell dissociation solution containing 0.02% (w/v) tetrasodium EDTA (EDTA-Na₄, 0.48 mM) in calcium- and magnesium-free D-PBS. This product is phenol red-free, animal component-free, and chemically defined.
EDTA chelates divalent metal ions such as Ca²⁺ and Mg²⁺, weakening cell-to-cell and cell-to-matrix adhesion to facilitate gentle cell dissociation. Because it contains no proteases, EDTA helps preserve the integrity of cell-surface proteins, making this solution suitable for studies of cell-surface receptors, flow cytometry, immunostaining, and immunofluorescence. It is primarily used to dissociate weakly adherent cells. It can also be used to wash cells before trypsin treatment or in combination with trypsin as needed. Use of this solution alone is not recommended for routine passaging of strongly adherent cells.
Protocols
1. Before use, allow an appropriate volume of Versene Solution to reach room temperature or warm it to 37°C.
2. Aspirate the culture medium and wash the cells 1–2 times with sterile Ca²⁺- and Mg²⁺-free PBS (MCE, Cat. No.: HY-K3005) or D-PBS (MCE, Cat. No.: HY-K3008).
3. Add enough Versene Solution to completely cover the cell layer. A recommended volume is 1.5 mL per 25 cm² of culture surface area. Incubate at room temperature for 1–2 min; extend the incubation if necessary.
4. When the cells shrink and round up and the spaces between them increase, gently tap the culture vessel or pipette gently to detach and collect the cells.
5. Centrifuge under conditions appropriate for the cell type. Discard the supernatant and resuspend the cells in culture medium or buffer for subsequent experiments.
6. If cell dissociation is incomplete, add more Versene Solution and extend the incubation time as needed. Avoid excessive treatment.

Attention
1. This product has been sterilized by filtration. Use aseptic technique to avoid contamination.
2. This product provides very gentle cell dissociation and is not recommended for routine passaging of strongly adherent cells.
3. Because EDTA is not neutralized, wash the detached cells once with PBS to remove residual EDTA, which may otherwise interfere with cell attachment.
4. This product is intended for scientific research by trained personnel only. It must not be used for clinical diagnosis or treatment, or in food or pharmaceutical products.
5. For your safety and health, wear a lab coat and disposable gloves when handling this product.
