Cytotoxic T lymphocyte-associated antigen-4 (CTLA-4, also known as CD152), a member of the immunoglobulin superfamily (IgSF)+, is a inhibitory receptor expressed exclusively on T cells and plays a critical role in the priming phase of the immune response. CTLA-4 is a CD28 homolog that binds CD80 (B7-1) and CD86 (B7-2) with a higher affinity than CD28. Therefore, CTLA-4 regulates the degree of co-stimulation by competing for binding. Blockade of CTLA-4 in vitro and in vivo can enhance T-cell proliferation and effector functions. CTLA-4 is the first immune-checkpoint receptor to be clinically targeted.
