Description & Advantages

SDS-PAGE Sample Loading Buffer (4×) is appropriate for loading protein samples on SDS-PAGE. The Buffer contains the coomassie brilliant blue (CBB) G250 and Tris (2-carboxyethyl) phosphine hydrochloride (TCEP, DTT Substitute), without Dithiothreitol (DTT) and 2-Mercaptoethanol. The migration speed of proteins is inversely proportional to its molecular weight, the smaller the molecular weight, the faster the migration speed.

プロトコル

1. Completely thaw SDS-PAGE Sample Loading Buffer (4×) and mix thoroughly before use.

2. Mix 3 parts protein sample with 1 part 4× Loading Buffer to obtain a final 1× loading buffer concentration.

3. Mix thoroughly and heat at 95-100°C for 5-10 min to denature the proteins.

4. Briefly centrifuge to collect the solution at the bottom of the tube.

5. Load an appropriate amount according to protein concentration and well capacity for SDS-PAGE.

Note: The buffer contains the reducing reagent TCEP and tracking dye. No additional DTT or β-mercaptoethanol is required.
