Labeling and Fluorescence Imaging

The click chemistry strategy can further and accurately study the biochemical state of cells and their small molecule perturbations. Bioorthogonal reactions allow for the incorporation of chemical groups with highly selective reactivity into small molecules, peptides, sugar chains, proteins, or nucleic acids modifications without affecting their biological function, enabling the selective installation of an analysis tag for downstream investigations. One of the most interesting functions of click chemistry is fluorescence imaging of intracellular target proteins (TOI). In the iEDDA reaction, innate TOI proteins in living cells can be visualized through the treatment of a TCO-ligand conjugate and Tz containing fluorophores (FLTz).
