MCE Exosome-Specific 293 Cell Medium Supplement is designed as a feed supplement for use with the Exosome-Specific 293 Cell Medium (Serum-Free) (MCE Cat. No.: HY-K3113). It is suitable for the development and production of exosomes derived from HEK293 cells. When used in combination with the Exosome-Specific 293 Cell Medium (Serum-Free), this supplement eliminates the need for serum during cell culture and significantly enhances exosome secretion by 293 cells. Compared to conventional HEK293 culture media, exosome yield can be increased by 5 to 10-fold.
Protocol
1. Expand the HEK293 cells to a density of 6 × 106 - 8 × 106 cells/mL.
2. Add the supplement at 25% of the culture volume. Continue culturing for 6-16 h, and collect the culture supernatant before cell viability drops below 90%.
3. The collected culture supernatant can be used for exosome isolation and purification.
